Research Interests: Synapses are modulated in ways that are critical for the normal functioning of the mammalian brain. The strengthening and weakening of synaptic transmission is known as synaptic plasticity. The overall goal of my research program is to identify and describe the mechanisms that control short-term and long-term synaptic plasticity using whole cell patch-clamp recordings in acute brain slices. Short-term synaptic plasticity, which last for seconds, may be important for different types of brain functioning and may be mediated by different cellular and molecular mechanisms then long-term synaptic plasticity. However, many aspects of both short-term and long-term synaptic plasticity remain poorly understood and few presynaptic mechanisms have been identified.Schizophrenia is a severe and chronic mental illness characterized by the onset of disorganized thought and speech in late adolescence and diagnosed by the persistent appearance of two or more dehabilitating symptoms including hallucinations, delusions, disorganized speech, catatonia, apathy and anhedonia or a flattening of affect. Functional neuroimaging and postmortem studies have revealed a number of abnormalities in the functioning of the prefrontal cortex (PFC) of schizophrenics. For example, dendritic spine density is substantially diminished in PFC neurons of patients with schizophrenia. The PFC plays an important role in many cognitive processes and is significantly affected in schizophrenia. A decrease in spine density may reflect a decrease in synaptic transmission and neuronal connectivity in the PFC and may cause the cognitive abnormalities observed in schizophrenics. Recently my research has focused on how trans- synaptic EphB/ephrinB signaling plays an important role in controlling both presynaptic neurotransmitter release and postsynaptic clustering of NMDA-receptor and/or AMPA receptor subunits in the PFC. My lab is currently using Golgi-Cox staining combined with patch clamp based synaptic physiology to study how EphB receptors may be critical for the development and maintenance of dentritic spines and how forward and reverse signaling may regulate synaptic transmission and neuronal connectivity in the PFC.

Research Interests: Administration of bright light is used as a treatment for seasonal depression. Although seasonal depression occurs in only a small percentage of the population, the majority of the population outside tropical and sub-tropical latitudes is aware of some seasonal changes in mood and behavior. Some, but not all studies found an improvement of mood in healthy people with mild seasonality when light is administered in winter. Light is thought to act in part by increasing serotonin synthesis and increasing serotonin synthesis by giving its precursor tryptophan can not only improve mood but also decrease quarrelsome behaviors and increase agreeable behaviors in some healthy people. The interest of our lab is to test whether bright light administered in winter can cause similar improvements in social behavior in healthy people with mild seasonality.

Research Interests: My primary interest is in the application of neuroimaging techniques to study the biological basis of psychopathology and related vulnerabilities. My past work used both PET dopamine imaging and fMRI to study populations at increased risk of schizophrenia by examining stress-related differences in dopamine transmission and activation of dopamine circuits of these populations. I have extended my work to examining the effects of stress on monoamine oxidase A in healthy and depressed individuals, while also examining neuroendocrine and personality variables. Another of my interests is the contribution of personality to the biology of vulnerability. I hope to describe the neurochemical correlates of vulnerable personality types using PET imaging. I hope this work will contribute to a better understanding of some of the physiological and environmental vulnerabilities to mental illness.

Research Interests: We have previously identified a heteromeric D1-D2 dopamine receptor signaling complex that is coupled to Gq-protein mediated intracellular Ca2+ release. I am now interested in studying the mechanisms regulating the D1–D2 heterooligomer signaling cascade. While there is much data on the regulatory profiles of the D1 and D2 receptors individually, there is little information on how the D1 and D2 receptors desensitize when co-expressed. Since desensitization/internalization operates as a safety device to prevent over-stimulation of receptors in the face of excessive agonist exposure, it is of interest to investigate and characterize the regulatory mechanisms of this novel signaling complex by using specific D1 and D2 agonists as pharmacological tools. Differences in the effect of D1 or D2 receptor selective agonists on the calcium signal mediated by the D1-D2 receptor complex may have relevance for clinical conditions where these selective agonists are used therapeutically. Continued investigation of this unique signal may yield a better understanding of behavioral and central nervous system disorders mediated by dopamine such as drug addiction and schizophrenia.
